Astract: Turtle live in tropical and subtropicaFwaters.All-mafine turtle spei:les-are m endangered category. One of turtle species knawn in the world is green turtle. Considering the collapse condition of turtle in Indonesia, insist conservation attempt to protect from decreasing. Genetic information became important to support conservation in the field. Goal of this research was to obtain genetic information of nucleotide base which is position, number and frequency of variable sites in turtle control region mtDNA sequences of Chelonia mydas from one mother in one eggs laying, as an establish step to observe genetic variation. Sample was taken from flipper tissue, isolating the DNA, amplified with PCR then sequencings the PCR product. Result shawed 34 variable sites in turtle control region mtDNA sequence. Data was analyzed by Sequence Alignment method and referred to the turtle control region sequence of Chelonia mydas from GenBank.
